You can take the train to Siena, then the bus to GAiole. www.trenitalia.com, Bus Number 127 from Siena to Gaiole.

Or train to Montevarchi and a bus? However, how are you going to get around once you arrive in Gaiole? The town itself is OK but the real interest in this area is the countryside, castles, vineyard estates and for that you'll need a car. Unless, of course, you are coming on cycling holiday, Gaiole always appearing to me to be the cycling capital of central Italy.
