Easiest is to get a car service like Dial7 to pick you up at the airport and take you to the hotel. It will be expensive since you'll need a large car for five of you, the base rate will be over $100 and you'll add the tip on top of that. That's for an outside pick-up: an inside pickup starts at $150.
The public transportation to your hotel is a single subway line from the airport monorail, if the kids can carry their own luggage. You'd take the AirTrain to Howard Beach (NOT Jamaica) and transfer to the Manhattan-bound A train to Jay Street station. Your hotel is just a few blocks from there. The AirTrain costs $5 per person, and the subway costs $2.75 per person or one swipe of an unlimited Metrocard per person (if you're buying them anyway), so significantly less than a car transfer.

If cost is a concern, there are some less expensive Brooklyn-based car services . . . I just checked the one I generally use (Arecibo), and their base rate between JFK and Park Slope is $53 ($38 plus an extra $15 for a van/SUV, which it sounds as though you'll need). Getting to the Sheraton in Downtown Brooklyn should be about the same cost as getting to Park Slope.
It's true that a Brooklyn based service will be cheaper, but I would still expect an additional $10-15 to downtown Brooklyn.
