Quickest/easiest and cheapest often don't go together. We went to Jiquilillo (taxi to UCA, minibus to Leon, bus to Chinandega, change bus stations, bus to Jiquilillo). This website has transportation options to Padre Ramos, including public transport. Apparently there are direct buses to Padre Ramos from Chinandega:
As Sonia mentioned easy, cheap and quick will be dificul, public buses will almost take forever but will be way much cheaper than private a private ride will be around US$130.00
Sonia's directions are great but there is another option too. By pass Leon by:
Take a cab to Mercado Israel where buses leave for Chinandega every 15 minutes. Travel time 1.5 hours
From Chinandega take a cab or tricycle to El Mercadito where buses leave for Jiquilillo/Padre Ramos at 7am, 10, 1130, 3pm, and 430 pm. Travel time is 1.5 hours.
Alternatively from Chinandega hire a cab for $20 to take you directly to Jiquilillo/Padre Ramos, travel time 45 minutes.
A direct cab from Managua to Jiquilillo/Padre Ramos is no more than $120 and takes about 2.5 - 3 hours.

Hey! Do you have info on reliable cabs to get to Padre Ramos from Managua Airport? Will they understand English or at least very basic Spanish/ google translate?also will cabs take US dollarsor only Corbodas?
