Being in Amsterdam without seeing the Van Gogh is a lost day of your life. Also do not forget the extra exhibitions in the same museum. The last one Prints in Paris 1900 was great.

No, but you have big museum store right by VG museum. If you need to go in a NEMO museum shop (perfect for children gifts) you can go without ticket.

If you have a timed ticket for 3:30 entrance, what's your actual time window for entrance? i.e. how early and late can you enter before/after your assigned time.

You will not be allowed in early and can arrive 30 minutes or so late.
